On the Utility of Heteroplasmy in Genotoxicity Studies: An Example from Chornobyl
نویسندگان
چکیده
We examine the utility of mtDNA heteroplasmy in assessing genetic damage due to environmental insult. Site heteroplasmy was quantified in a 400 bp portion of the cytochrome b gene in voles from a contaminated area near Chornobyl, Ukraine and from a relative control site by examining pregnant females and their embryos. A four hundred base pair segment was sequenced from approximately ten clones from each mother and embryo. Taq andror cloning mutations were evaluated to estimate technical error. Although the rates of substitutions in clonal variants for experimentals, biological controls and technical controls were 1 in 1,840 bp, 1 in 2,280 bp, and 1 in 3,333 bp, respectively, we could not reject the null hypothesis that the variants were the result of a single, combined, mutation rate. However, multiple substitutions and transversions were restricted to clones from the Chornobyl samples. Bootstrap analyses indicate that these aspects of variation were significantly different from the controls. Examination of the mitochondrial genome by cloning individual molecules for site heteroplasmy to estimate effects of pollution on mutation rate in free-living organisms appears to warrant additional study.

Assessing the genotoxicity of chronic environmental irradiation by using mitochondrial DNA heteroplasmy in the bank vole (Clethrionomys glareolus) at Chornobyl, Ukraine.
This study was designed to investigate whether or not chronic exposure to Chornobyl radiation poses a molecular genetic risk to mammals by examining a relatively rapidly evolving genetic system, mitochondrial DNA (mtDNA). More mtDNA mutations (approximately 19%) and an increase in mtDNA heteroplasmy (approximately 5%) occurred in the cytochrome b gene of an exposed mother-embryo set when compar...

متن کاملTranslational Antidote Research: A Bedside to Bench Tale
Although antidote development should proceed in an orderly fashion from observation, to experimental and safety studies, to clinical trials, this sequence is not always precisely followed. The development of fomepizole as an antidote for toxic alcohol and glycol poisoning is an example of how this may not be the case. Interest in the development of fomepizole was spurred in the 1960s. Shortly t...

Background and Theoretical Foundations: Transportation is one of the necessities for the economic development of any country. Among various modes of transportation such as air, land and sea the sea transportation is the cheapest, and it is more important as the worlds continue to grow. In Iran, due to political issues and sanctions, and since imposing limitation on marine transport cannot be im...
